Whenever you do ANYTHING in the layout or in terms of things like fancy scripting, ask yourself "How does this make the site more useful to visitors" - if the answer is that it doesn't, LEAVE IT OUT! In particular flashy animated nonsense and endless "gee ain't it neat" scripttardery just gets in the way of what's important -- delivering content to users.
If you have a well organized fast loading accessible layout with content people are actually interested in, you'll do well regardless of what that content is.
